RESUMO

PURPOSE: XinJiaCongRongTuSiZiWan (XJCRTSZW) is a traditional Chinese medicine compound for invigorating the kidney, nourishing blood, and promoting blood circulation. This study aimed to explore the effect of XJCRTSZW on triptolide (TP)-induced oxidative stress injury. METHODS: Adult female Sprague-Dawley rats and human ovarian granulosa cell lines were treated with TP and XJCRTSZW. Hematoxylin and eosin staining, enzyme-linked immunosorbent assay, flow cytometry, CCK-8, JC-1 staining, transmission electron microscopy, reverse transcription-quantitative polymerase chain reaction, and Western blotting were performed in this study. RESULTS: XJCRTSZW treatment observably ameliorated the TP-induced pathological symptoms. Furthermore, XJCRTSZW treatment observably enhanced the TP-induced reduction of estradiol, anti-Mullerian hormone, progesterone, superoxide dismutase, ATP content, mitochondrial membrane potential, p62, and Hsp60 mRNA, and protein levels in vivo and in vitro (p < 0.05). However, TP-induced elevation of follicle stimulating hormone and luteinizing hormone concentrations, malondialdehyde levels, reactive oxygen species levels, apoptosis rate, mitophagy, and the mRNA and protein expressions of LC3-II/LC3-I, PTEN-induced kinase 1 (PINK1), and Parkin were decreased (p < 0.05). In addition, XJCRTSZW treatment markedly increased cell viability in vitro (p < 0.05). CONCLUSIONS: XJCRTSZW protects TP-induced rats from oxidative stress injury via the mitophagy-mediated PINK1/Parkin pathway.

RESUMO

Mammalian target of rapamycin (mTOR) is an evolutionary conservative serine/threonine protein kinase. Its biological function is mainly to participate in cell proliferation, growth and differentiation, so as to regulate the body's metabolic process. Many domestic and foreign studies have shown that mTOR is the junction of apoptosis and autophagy signal transduction pathways. Various stimuli, such as nutrition, drugs and oxidative stress, may play a key regulatory role in cell apoptosis and autophagy through mTOR-mediated signaling pathways. At present, many studies have shown that the change in mTOR signaling pathways is closely related to the pathogenesis of many human diseases, such as cancer, metabolic disorders(obesity and type 2 diabetes), cardiovascular and neurodegenerative disease, age-related diseases and disorders of follicular. In recent years, more and more doctors have studied the regulatory effect of traditional Chinese medicine on apoptosis and autophagy, with the pathways as the starting point and cell apoptosis and autophagy as the research carriers. These studies include experimental studies on the regulation of apoptosis and autophagy by monomers of traditional Chinese medicine (TCM), Chinese patent medicine, compound prescription of TCM, acupuncture and other drugs and physiotherapy. Starting from the mTOR signaling pathways, this paper discusses the relationship between mTOR, apoptosis and autophagy, and reviews the recent progress of TCM in regulating apoptosis and autophagy through mTOR pathways, so as to provide ideas and references for further studies in this field. In the future, TCM doctors can still explore the time-effect relationship between apoptosis and autophagy based on mTOR signaling pathways under the guidance of the basic theory of Chinese medicine, in order to provide theoretical support and targets for the action mechanism of TCM on bodies.

RESUMO

The aim of this paper was to observe the concentration,time and mechanism of autophagy induced by triptolide( TP) in ovarian granulosa cells( OGCs). CCK-8 method was used to compare the inhibitory effects of TP at different concentrations on primary cultured rat OGCs and IC50 was calculated. The effects of TP at different concentrations and time points on the expression of OGCs autophagy factor protein and the cascade of PI3 K/AKT/m TOR pathway were detected by Western blot. The effects of TP,autophagy inducer( brefeldin A) and PI3 K/m TOR inhibitor( NVP-BEZ235) on the expression of PI3 K/AKT/m TOR cascade and autophagy related factor protein were detected by Western blot. The results show that the IC50 of different concentrations of TP on OGCs of rat ovary was14. 65 μmol·L-1,and the minimum inhibitory concentration of TP was 0. 1 μmol·L-1( 100 nmol·L-1). Compared with the control group,the expression levels of beclin1 and LC3Ⅱ in each group were significantly higher than those in the control group( P<0. 05 or P<0. 01). After 12 hours of treatment with TP,brefeldin A and NVP-BEZ235,respectively,compared with the control group,TP could significantly promote the expression level of downstream autophagy effect or molecule beclin1,LC3Ⅱ and inhibit the expression level of LC3Ⅰ,p62 protein( P<0. 05 or P< 0. 01). Moreover,the expression of beclin1 and LC3Ⅱ/LC3Ⅰ in TP group was higher than that in brefeldin A group( P<0. 05 or P<0. 01),and the expression of p62 in TP group was lower than that in brefeldin A group( P<0. 05 or P<0. 01). At the same time,TP could significantly inhibit the expression of p-PI3 K,p-AKT,p-mTOR protein,and the inhibitory effect of TP was better than that of NVP-BEZ235 group. This study suggests that 100 nmol·L-1 TP could induce OGCs autophagy successfully in cultured rat ovary for 12 h; TP may induce OGCs autophagy by inhibiting PI3 k/Akt/m TOR signaling pathway.

In the theory of traditional Chinese medicine(TCM) that "kidney storing essence and governing reproduction", reproductive essence is an important part of the kidney essence and acts as the original material of offspring embryos. Sperm, oocyte and zygote should be all included in the range of reproductive essence. Ovum is the essence of reproduction from inborn. The follicles maturation depends on the quality of oocyte and the vigor of kidney essence. Meanwhile, discharge of mature ovum relies on the stimulation and promotion by kidney Qi. Autophagy almost exists in different cells stages and all various of mammalian cells. Many studies have found that autophagy not only participates in the formation of follicles, but also in every phase of the follicles development, and is involved in the occurrence and development of ovarian diseases. Recently, more and more scholars believe that autophagy is a new field to explore the microcosmic relationship between autophagy and TCM. Kidney-nourishing TCM could promote follicular growth and improve variety clinical symptoms by inhibiting the apoptosis of ovarian granulosa cells and reducing follicular atresia. Meanwhile, apoptosis of ovarian granulosa cells is closely related to autophagy of ovarian granulosa cells. In order to provide some theoretical foundation for kidney-nourishing therapy's promoting effect on follicular growth and improving effect on ovarian function, also to further explore the molecular mechanism of kidney-nourishing medicine in promoting follicular development, this paper would explain the microcosmic relationship between autophagy and follicular development based on the theory of "kidney governing reproduction". All of these would be of great significance to prevent and intervene the diseases of reproductive system timely and effectively.

RESUMO

The factors impacting the pregnancy rate and the live birth rate mainly include ovary function disorder and low endometrial receptivity, which can cause the difficulty in embryo implantation, early miscarriage and pregnancy failure. In recent years, researchers of the traditional Chinese medicine (TCM) have made active efforts in assisting IVF-ET, so as to achieve a great advance in improving the ovary reaction, treating the ovarian hyperstimulation syndrome (OHSS), improving the follicle, embryo quality and endometrial receptivity and protecting the fetus, which had been summarized in this article.

RESUMO

[ Objective] To investigate the effect of community -based comprehensive intervention on improving residents'knowledge , attitude and behaviors in terms of stroke prevention . [ Methods] By means of stratified sampling , 120 resident population were randomly selected from Xietu Neighborhood of Shanghai .Demographic characteristics , prevalence of stroke , knowledge , attitudes and behaviors related to stroke prevention were investigated through baseline survey .After 3-month interventions , end stage survey was conducted to assess the effects of the interventions by measuring the changes of the related indicators as mentioned above . [ Results] With intervention , the residents'knowledge and attitude on stroke preven-tion was raised .The number of the residents was increased participating in prevention , education and coun-seling activities on stroke prevention and those who voluntarily went to hospital for early screening on stroke were increased , too. [ Conclusion] Community-based comprehensive interventions could be effective in improving residents'knowledge , attitudes and behaviors on stroke prevention .Conducting community in-tervention on stroke is necessary .
